Researchers from the University of Nebraska Medical Center published a landmark study in Molecular Psychiatry analyzing over six million maternal-child health records from 2014-2023, representing one-third of all U.S. births during that period. The study found that prenatal exposure to sterol biosynthesis-inhibiting medications—including common antidepressants like fluoxetine and sertraline, beta-blockers like propranolol, and statins—increased autism spectrum disorder risk by 1.47-fold for one medication and 2.33-fold for four or more co-prescribed drugs. Exposure to these medications during pregnancy surged from 4.3% in 2014 to 16.8% in 2023, affecting growing numbers of pregnancies. The researchers linked the findings to disruption of cholesterol production critical for fetal brain development around 19-20 weeks of gestation, drawing parallels to Smith-Lemli-Opitz syndrome where 75% of affected children exhibit autism symptoms. While advocating for heightened clinical vigilance and integration of sterol metabolism assessments into drug development, researchers emphasized that patients should not abruptly stop prescribed treatments without medical guidance.
